RESUMEN

Preeclampsia (PE) is a serious complication, and soluble fms-like tyrosine kinase (sFLT1) released from the placenta is one of the causes of PE pathology. Trophoblasts are the primary source of sFLT1; however, monocytes/macrophages exist enough in the placenta can also secrete sFLT1. Sterile inflammatory responses, especially NLRP3 inflammasome and its downstream gasdermin D (GSDMD)-regulated pyroptosis, may be involved in the development of PE pathology. In this study, we investigated whether human monocyte/macrophage cell line THP-1 cells secrete sFLT1 depending on the NLRP3 inflammasome and GSDMD. To differentiate THP-1 monocytes into macrophages, treatment with phorbol 12-myristate 13-acetate (PMA) induced sFLT1 with interleukin (IL)- 1ß, but did not induce cell lytic death. IL-1ß secretion induced by PMA inhibited by deletion of NLRP3 and inhibitors of NLRP3 and caspase-1, but deletion of NLRP3 and these inhibitors did not affect sFLT1 secretion in THP-1 cells. Both gene deletion and inhibition of GSDMD dramatically decreased IL-1ß and sFLT1 secretion from THP-1 cells. Treatment with CA074-ME (a cathepsin B inhibitor) also reduced the secretion of both sFLT1 and IL-1ß in THP-1 cells. In conclusion, THP-1 macrophages release sFLT1 in a GSDMD-dependent manner, but not in the NLRP3 inflammasome-dependent manner, and this sFLT1 release may be associated with the non-lytic role of GSDMD. In addition, sFLT1 levels induced by PMA are associated with lysosomal cathepsin B in THP-1 macrophages. We suggest that sFLT1 synthesis regulated by GSDMD are involved in the pathology of PE.

RESUMEN

A computational technique based on the lattice Boltzmann method (LBM) is developed to simulate the wettable particles adsorbed to a liquid-vapor interface under gravity. The proposed technique combines the improved smoothed-profile LBM for the treatment of moving solid particles in a fluid and the free-energy LBM for the description of a liquid-vapor system. Five benchmark two-dimensional problems are examined: (A) a stationary liquid drop in the vapor phase; a wettable particle adsorbed to a liquid-vapor interface in (B) the absence and (C) the presence of gravity; (D) two freely moving particles at a liquid-vapor interface in the presence of gravity (i.e., capillary flotation forces); and (E) two vertically constrained particles at a liquid-vapor interface (i.e., capillary immersion forces). The simulation results are in good quantitative agreement with theoretical estimations, demonstrating that the proposed technique can reproduce the capillary interactions between wettable particles at a liquid-vapor interface under gravity.

Cryptosporidium is a common intestinal protozoan that can lead to diarrhea in humans and dogs. The predominant species of infection are C. hominis and C. parvum in humans, and C. canis in dogs. However, C. canis can infect immunocompromised humans. Considering the close contact with humans, dogs have the potential to be reservoirs for human cryptosporidiosis. Breeding kennels are the major supply source of puppies for pet shops. The present study is to determine the molecular prevalence and characteristics of Cryptosporidium spp. found in breeding kennel dogs. A total of 314 fecal samples were collected from young and adult dogs kept in 5 breeding kennels. A polymerase chain reaction targeting the small subunit rRNA gene was employed for the detection of Cryptosporidium spp. To determine the species, the DNA sequences were compared to GenBank data. Overall, 21.0% of the fecal samples were positive for Cryptosporidium spp. infection. Cryptosporidium spp. was detected in all 5 facilities. A sequencing analysis demonstrated that all isolates shared 99-100% similarity with C. canis. The results suggest that Cryptosporidium spp. infection is present at a high-level in breeding kennel dogs. However, because dominant species in this survey was C. canis, the importance of breeding kennel dogs as reservoirs for Cryptosporidium spp. transmission to humans is likely to be low in Japan.

PROBLEM: We investigated the effect of palmitic acid (PA), a major saturated fatty acid in NEFA, on bovine oviduct epithelial cells (OECs) during in vitro cell culture. METHOD OF STUDY: Bovine oviductal tissues ipsilateral to the corpus luteum were collected 1-3 days after ovulation; the OECs were isolated and cultured. RESULTS: PA increased lipid accumulation and activated caspase-3 in OECs, resulting in decreased cell proliferation. PA also stimulated the secretion of inflammatory cytokine interleukin (IL)-8 depending on TLR4, NF-κB activation, and reactive oxygen species (ROS) production. Moreover, PA induced mitochondrial dysfunction, including mitochondrial fission, ATP production, and mitochondrial ROS production. It also increased levels of LC3 and p62 proteins, suggesting autophagy induction in OECs. CONCLUSION: We suggest that bovine OECs recognize an excessive increase in endogenous and sterile danger signals, such as PA, which may contribute to chronic oviductal inflammation, resulting in infertility associated with oviductal dysfunction.

The mammalian oviduct is an essential site for sperm storage, the transport of gametes, fertilization, and embryo development-functions that are aided by cytokines secreted from oviduct epithelial cells (OECs). Aging leads to cellular and organ dysfunction, with infertility associated with advanced maternal age. Few studies have investigated age-dependent changes in the oviduct as a possible cause of infertility, so we compared OECs from young (30-50 months) versus aged (more than 120 months) cattle. Next-generation sequencing was first used to identify age-related differences in gene expression. Several proinflammatory-related genes (including IL1B, IL1A, IL17C, IL8, S100A8, S100A9, and TNFA) were activated in OECs from aged (more than 120 months) compare to young (30-50 months) individuals, whereas genes associated with extracellular matrix-related factors (COLs, POSTN, BGN, and LUM) were down-regulation in aged OECs. Indeed, IL1 B and IL8 abundance was higher in aged OECs than in young OECs. Young OECs also tended to proliferate faster, and the revolution frequency of young, ciliated OECs was higher than that of their aged counterparts. In contrast, aged OECs possessed more F-actin, an actin cytoskeleton marker associated with reduced elasticity, and contained high levels of reactive oxygen species, which are mediators of inflammation and senescence. These different functional characteristics of bovine OECs during the post-ovulatory phase support the emerging concept of "inflammaging," that is, age-dependent inflammation. Mol. Reprod.
